All harmonica lessons I have seen say that the harmonica should be held with the numbers facing up. This feels very awkward to me and I always seem to be more comfortable with the the numbers facing down. Does it really matter?

Generally speaking, no worries, you'll be fine... just reverse all of the directions in regards to tongue blocking (tongue goes to the right for standard tongue blocking techniques... like slaps and flutters for holes 2-10 and to tongue goes to left for hole 1, if you decide to tongue block hole 1).
One challenge is cupping. When holding the harmonica with your left hand, the cup created is focused on the commonly-used low end of the harmonica. In your case this will be reversed, compromising your Wa Wa effects and amplified tone. I suggest you study and use the "Two Thumbs Up" approach I teach in the "Cupping & Hand Effects" lesson.
