While you can check tuning against a reference plate, tuning is always best tested in assembled state, as several thigns about the assembled state may influence tuning, such as the shape of the reed chamber, aky air flow passing around the other reed in the chambe,r and the influence of covers on pitch.
Start by plinking hte reed several times, then assemble the comb with both reedplates. adding the covers and applying finger pressure on the covers will give sufficient airtightness for testing purposes.
